Two people were killed as their car was fired at near the NIPA Chowrangi flyover.
Police said the men identified as Moulana Shahabullah and his guard Sanober Khan were critically injured and were shifted to a hospital where they succumbed to their wounds.
A passer-by was injured as the car hit him during the attack, police added.
Moulana Shahabullah was en route to a mosque to lead Friday prayers when his car came under attack, officials said.
Four assailants on two motorcycles fired at the car from both sides and fled, police said.
It is believed to be an incident of targeted killing, the officials further stated.
Sindh Chief Minister Murad Ali Shah took notice of the incident and sought a report from Additional IG Karachi.
The Sindh chief minister asked the additional IG to specify whether it is an incident of targeted killing or not. Shah also ordered the immediate arrest of the suspects.
Separately, one person was killed and another injured in firing at a car on Shahrah-e-Faisal near Nursery.
